ABSTRACT

Objective To investigate the differences in genotypes and phenotypic parameters of β-thalassemia gene carriers in pregnant women′s from Chengdu, Sichuan Province.Methods Totally 320pregnant women′s withβ-thalassemia gene from March 2016to June 2017in our hospital were selected.Routine blood tests, alkaline hemoglobin electrophoresis and routine analysis ofβ-thalassemia were performed on all the cases.Statistical analysis was performed on the data of each group.Results There were 306cases of heterozygousβmutations and 10types of mutations, among which 14cases ofα-thalassemia combined had 6types of mutations.The mutations of MCV, MCH, MCHC, and Hb in the routine blood tests of each group showed some differences.The incidence of abnormal bands was also different for each mutation, and the hemoglobin electrophoresis results ofβEM mutations contained abnormal bands.However, the clinical manifestations of CAPM mutations were not obvious and easily missed.Conclusion There is a certain regional specificity inβthalassemia gene carrying in Chengdu area.Targeted examination in the preliminary screening and prenatal diagnosis should be conducted so as to reducing the birth rate of children′s with severe thalassemia.

ABSTRACT

Objective To investigate the regulatory effects of miR-145 on MMP2 expression in lung adenocarcinoma cell A549 and the mechanism involved in the regulation.Methods Lipofectamine 2000 reagent-mediated miR-145 mimic or mimic negative control (NC) was transiently transfected into lung adenocarcinoma cell A549.The expression levels of miR-145 were determined by RT-PCR.MMP2 and β-catenin expression was detected by western blot.Lithium chloride (LiC1),the activator of Wnt/β-catenin pathway,and miR-145 mimic co-processed to act on A549 cells,then the expressions of MMP2 and β-catenin were detected by Western blot.Results The results of RT-PCR showed that the difference of miR-145 in miR-145 mimic group,NC group and blank group was statistically significant(F =296.30,P <0.05).western blot showed that the expression levels of MMP2 and nucleus β-catenin were significantly downregulated in miR-145 overexpression group compared with the control (q =8.98,26.59,all P < 0.05).Additionally,the effects of co-processing factors on the cells showed that the activation of wnt/β-catenin pathway reversed the inhibition of miR-145 on MMP2 expression(q =49.47,P < 0.05).Conclusion miR-145 may downregulate MMP2 expression through inhibition of Wnt/β-catenin pathway activity in lung adenocarcinoma cell A549.

ABSTRACT

Objective To investigate the application of plasma creatine kinase isoenzyme (CK-MB) ,N-ter-minal pro-brain natriuretic peptide(NT-proBNP)and myocardial nutrient-1(CT-1)in the diagnosis of neonatal asphyxia complicating myocardial injury .Methods Eighty cases of neonatal asphyxia in the hospital from Jan-uary to December 2016 were selected ,including 28 cases in the asphyxia non-myocardial injury group ,32 cases in the mild asphyxia myocardial injury group and 20 cases in the severe asphyxia myocardial injury group .Oth-er 80 healthy neonates were selected as the control group .The levels of plasma CK-MB ,NT-proBNP and CT-1 on 1 ,3 ,5 d after birth were measured by adopting the ELISA method .The relationship between plasma CK-MB ,NT-proBNP and CT-1 levels with prognosis in neonatal asphyxia was analyzed .Then the application value of plasma CK-MB ,NT-proBNP and CT-1 in the prognosis evaluation of neonatal asphyxia complicating myo-cardial injury was analyzed by using the receiver operating characteristic (ROC) curve .Results Plasma NT-proBNP ,CK-MB and CT-1 levels in the control group had no statistically significant difference among 1 ,2 ,3 d after birth (P>0 .05) ,but plasma CK-MB ,NT-proBNP and CT-1 levels on postnatal 3 d in the neonatal as-phyxia group reached the peak value ,plasma CK-MB ,NT-proBNP and CT-1 level on postnatal 1 ,3 d in the se-vere asphyxia myocardial injury group and mild asphyxia myocardial injury group were significantly higher than those in the non-myocardial injury group and control group ,and w hich in the non-myocardial injury group were higher than those in the control group ,the differences were statistically significant (P<0 .05) .Plasma NT-proBNP ,CK-MB and CT-1 levels on postnatal 5 d in the non-myocardial injury group and mild asphyxia myocardial injury group returned to normal levels ,w hile w hich in the severe asphyxia myocardial injury group were still higher than those in the other groups ,the differences were statistically significant (P<0 .05) .The levels of plasma CK-MB ,NT-proBNP and CT-1 in the death neonates with neonatal asphyxia were higher than those in the survival neonates ,and the difference was statistically significant (P<0 .05) .The sensitivity and specificity of combined detection of plasma CK-MB ,NT-proBNP and CT-1 for the prognosis diagnosis in neo-nates with asphyxia complicating myocardial injury were higher than that of single index .Conclusion The early determination of plasma CK-MB ,NT-proBNP and CT-1 levels will be helpful to effectively evaluate the myocardial injury severity and prognosis in asphyxia neonates and has an important clinical value .

ABSTRACT

Objective To investigate and analyze the diagnostic significance of humoral and cellular immune function detection in the patients with syphilis.Methods Eighty-six cases of syphilis without anti-syphilis treatment in our hospital were selected as the research subjects(syphilis),including 26 cases of primary syphilis,39 cases of secondary syphilis 21 cases of latent syphilis,and 30 healthy subjects were selected as the control group.Then the humoral immunity and cellular immunity levels were compared between the two groups.Results The levels of IgG,IgA and IgM in the syphilis group were significantly increased compared with the control group(P0.05),which in the patients with secondary syphilis and latent syphilis were significantly increased compared with the control group,moreover the increase in the secondary syphilis was most significant(P<0.05);the CD3+,CD4+,CD4+/CD8+ and NK levels in different stages of syphilis were significantly decreased compared with the control group(P<0.05),while the CD8+ level was significantly higher than that in the control group(P<0.05),in which the change in the secondary syphilis patients was most significant.Conclusion The humoral immune in the patients with syphilis is hyperfunction,while cellular immune is dysfunction,the patients present the overall immune function disorder,therefore the humoral and cellular immunity detection has a certain clinical value in the diagnosis of syphilis.
